[225Ac]Ac-mcp-IgG4 is a radiolabeled isotype control antibody utilized in preclinical radioimmunotherapy research. It consists of a non-specific human or murine immunoglobulin G4 (IgG4) monoclonal antibody conjugated to the alpha-emitting radioisotope Actinium-225 ($^{225}$Ac) via the macrocyclic chelator MACROPA (mcp). In the context of targeted alpha therapy studies, such as those evaluating agents against Delta-like ligand 3 (DLL3), this construct serves as a negative control to differentiate the therapeutic efficacy of target-specific binding from the non-specific effects of systemic radiation and the pharmacokinetic behavior of the IgG4 scaffold. Actinium-225 provides high-linear energy transfer (LET) alpha particles that induce complex DNA double-strand breaks, while the MACROPA chelator ensures stable complexation of the radionuclide to the antibody.
